Role of Intracellular Ca2+ in the Diazoxide-induced Stimulation of Melanin Synthesis in B16 Melanoma Cells

Abstract

|
The effect of diazoxide on melanogenesis was investigated in B16 melanoma cells. Diazoxide stimulated the production of melanin in a dose-dependent manner. Diazoxide increased intracellular Ca2+ concentration, and BAPTA/AM significantly inhibited the diazoxide-induced melanogenesis. Diazoxide-induced intracellular Ca2+ increase and melano- genesis was significantly inhibited by EGTA. Taken together, these results suggest that intracellular Ca2+ influx may medi- ate the diazoxide-induced melanogenesis, and that diazoxide may be useful in the management of pigmentation disorder.
